1. Tuberculosis Antibody Diagnostic ELISA Kit Introduction

This kit adopts the indirect ELISA method to coat the Mycobacterium tuberculosis antigen on the enzyme microporous strip. During the detection, add the diluted serum sample. After incubation, if there are Mycobacterium tuberculosis-specific antibodies bound to the coating antigen, the unbound antibodies and other components are washed and discarded; The complexes with pre-coated antigens are bound; the unbound enzyme conjugates are discarded by washing; TMB substrate is added to the microwells to enzymatically form blue products, after adding stop solution to stop the reaction, use a microplate reader at 450nm The absorbance A value of the reaction well was measured at the wavelength.

3. Test procedure

1) Take the tuberculosis-silver-coated plate (open and measure the required amount according to the sample), add the diluted serum to the test wells, 100ul/well; at the same time, set 2 wells of positive control and 1 well of negative control, respectively in their wells Add negative control and positive control, 100ul/well (no overflow);
2) Cover with self-adhesive foil and incubate at 37°C for 30 minutes;
3) Open the film, discard the well solution, add diluted washing buffer to each well, 250ul/well, discard the solution, repeat the above steps 4-6 times, and finally pat dry with absorbent paper;
4) Add Enzyme Conjugate, 100ul/well, shake gently, cover with plastic wrap, and incubate at 37°C for 30 minutes;
5) Open the self-adhesive foil, pour out the liquid in the well, add diluted washing buffer to each well, 250ul/well, pour out the liquid, repeat the above steps 4-6 times, and finally pat dry with absorbent paper;
6) Add substrate, 100ul/well, mix well, cover with plastic wrap, and incubate at 37°C for 10 minutes in the dark;
7) Add stop solution, 50ul/well to stop the reaction, and measure the result in 10 minutes.

Read the OD value at 450nm (630nm as reference).
For the detection to work:
Negative control OD value (N)<0.2, while positive control OD value (P)>0.4
Calculation method:
S/P value = sample OD value/positive control OD average
Interpretation of results
S/P value < 0.3: negative
