1. Classical Swine Fever Virus Antibody Diagnostic ELISA Kit Principle
GreenSpring® Classical Swine Fever Virus (CSFV) Antibody ELISA Detection Kit is made from antigen-coated microtiter plates (CSFV antigen-coated) and other reagents. It uses the principle of solid-phase ELISA to detect CSFV-Ab in serum, and then adds an enzyme conjugate to specifically bind to the complex of coated antigen + CSFV-Ab + enzyme-labeled anti-pig IgG antibody on the microplate. With TMB substrates, a certain amount of color is produced. The depth of color is related to the content of CSFV-Ab. When the color value is greater than the critical value, the pig is well inoculated or naturally infected.

3. Results

For the assay to be valid, the mean OD value of the positive control wells must be greater than or equal to 1.0, and the mean OD value of the negative control wells must be less than 0.1. Otherwise, the test is invalid and needs to be re-tested.
The result is judged by the S/P value,
S/P=(sample OD450/630- NCx(—))/( PCx(—)- NCx(—)), NCx(—) represents the average OD450/630 value of the negative control (less than 0.05, calculated as 0.05), PCx(— ) represents the mean OD450/630 value of the positive control
S/P≥0.25 is positive; less than 0.25 is negative.

4. User Notices and Warnings

1. This kit is for research use only.
2. Do not use expired reagents, and do not mix reagents from different batches.
3. Please read the manual carefully before use.
4. The experimental garbage should be sterilized by high pressure steam at 121℃ for 30 minutes, or treated with 5.0g/L sodium hypochlorite disinfectant for 30 minutes and then discarded.
5. Microplates removed from refrigeration should be equilibrated and allowed to dry at room temperature before opening. Return unused MicroWell plates to dry foil bags and seal at 4 °C. Unused liquid reagents should be covered and stored together with other components at 2-8°C away from light.
6. Micropipettes should be used to add samples and reagents and should be frequently demonstrated for accuracy.
7. When adding washing buffer, it should be topped up but not overflowed to avoid free enzyme or cross-contamination between wells.
8. The stop solution is corrosive, immediately rinse with plenty of water when it comes into contact with skin or clothes.

Harmful symptoms

- The most acute type:

It is mostly seen in the pig farms at the beginning of the epidemic and the first occurrence of swine fever. The incubation period is generally 2-3 days. Pigs suddenly come on, high fever persists (body temperature rises by more than 2 ℃), systemic convulsions, limb convulsions, cyanosis of limb ends, ear tips and mucous membranes, multiple bleeding spots or spots on the whole body, unable to rise on the ground, and soon die. The course of disease is generally less than 5 days, and the mortality rate is 90-100%.

- Acute type:

The most common type in clinical practice, the incubation period is generally 3-5 days. At the beginning, the infected pigs are depressed and their appetite decreases sharply. At the later stage, the sick pigs are anorexic. They often put their mouth in the trough for a moment and return to the rest place. High fever persists, while the number of white blood cells decreases. At the initial stage of the disease, the secretion of the eyes of the sick pigs increased, and conjunctivitis accompanied by tears, and in severe cases, the eyelids were completely adhered. At the initial stage of high temperature, constipation usually occurs, followed by severe yellow-brown watery diarrhea and vomiting, and yellow bile containing liquid is vomited. Sick pigs are afraid of cold, often gather together, shiver, drowsy and fluffy. The sick pigs became thin and weak within a few days. Most of the pigs walked in a crooked or wobbly way. Their hind legs were paralyzed. They could not stand and sat in a dog position. They occasionally twitched. There are often red or purple bleeding spots on the abdominal skin, ear mirror, limbs, and even the whole body, gradually expanding into patches or bleeding spots, and even skin necrosis areas. Some diseased pigs' ear tips and tails changed from red to purple or even blue-black due to bleeding and necrosis, and gradually dried up, often being bitten by other pigs. Most pigs with acute swine fever died 10-20 days after infection, with a mortality rate of 50-60%. The body temperature dropped below normal hours before death.
